Yale Researchers Uncover Mechanism of Coenzyme A Transport into Mitochondria
Yale scientists have identified the transport system that moves Coenzyme A into mitochondria, shedding light on a crucial aspect of cellular metabolism and potential disease treatments.
Yale researchers have identified the mechanism by which coenzyme A (CoA), a molecule derived from vitamin B5 essential for metabolism, is transported into mitochondria. Their study, published in Nature Metabolism, reveals that specific transport systems are responsible for moving CoA into these cellular powerhouses.
CoA is predominantly found inside mitochondria, with up to 95% concentrated within these organelles. However, the process by which CoA reaches mitochondria was previously unclear. The research team developed a method using mass spectrometry to analyze CoA conjugates—compounds formed when CoA binds to other molecules. They identified 33 types of CoA conjugates in whole cells and 23 within mitochondria.
Further experiments indicated that the enzyme required to produce CoA is mainly located outside mitochondria. When researchers disrupted the molecular transporters responsible for moving CoA, mitochondrial CoA levels dropped significantly. These findings suggest that CoA is imported into mitochondria via specific transporters.
Understanding this transport mechanism could provide insights into diseases linked to CoA dysfunction. Mutations in genes encoding CoA transporters have been associated with encephalomyopathy, a condition involving developmental delays, epilepsy, and reduced muscle tone. Additionally, mutations in enzymes involved in CoA production have been linked to neurodegenerative diseases.
